Description
Description
DNA Polymerase is a thermostable instrument that is required for DNA amplification. DNA polymerase produces the necessary single-stranded DNA template for replication. Most thermostable DNA Polymerases have range of temperature from 70 to 74 . Some thermostable DNA Polymerases possess a reverse transcriptase activity which can be activated by using manganese instead of magnesium as a cofactor. DNA Polymerase can extend the annealed primers to produce primer dimer which often appears as a diffuse band of approximately 50 – 100 bp on an ethidium bromide-stained gel. The DNA Polymerase can be kept in an inactive state by binding to an oligonucleotide which is known as an aptamer, or through chemical modification. Many DNA Polymerases are supplied with a magnesium-free reaction buffer and a tube of 25 mM MgCl2 so that concentration of Mg2+ can be adjusted to the optimal level for each reaction. GoTaq® DNA Polymerase is a proprietary formulation of Taq Polymerase that gives robust amplification equal to and superior to that of standard Taq. The most commonly used DNA Polymerases for PCR have no reverse transcriptase activity under standard reaction conditions and thus amplification products will be generated only if the template contains trace amounts of DNA with similar sequences.
Features and Benefits:
- Revolutionized and popularized PCR: DNA Polymerase has ability to withstand the high denaturation temperatures.
- Improved the stringency of primer annealing: allow higher annealing temperatures.
- Convenience: direct to gel.
- Optimization: the 5X Green and 5X Colorless Reaction Buffers allow to go directly from thermal cycler to gel analysis.
- Efficiency: buffers contain a compound that increases sample density, so the sample sink easily into wells of an agarose gel.
- Easy monitoring during electrophoresis: green buffer that contain of two dyes (yellow and blue) that are separated. Blue dye comigrated with 3-5 kb DNA fragments in a 1% agarose gel while yellow dye migrates ahead of primers (<50 bp).
