WEBINAR Characterizing Molecular Glue Mediated Protein Degradation using Live-Cell Kinetic Methods, 14 July 2022
FREE PROMEGA WEBINAR
Date : 14th July 2022
Time : 11 am (Indonesia)
The ability to monitor degradation of endogenous protein levels amongst related classes, in live cells, is critical for understanding the efficacy and
mechanism of degrader compounds. In this webinar we will discuss the automation of parallel degradation and viability screening of neo-substrates using a panel of immunomodulatory drug (IMiD) molecular glues. We will show application of HiBiT CRISPR tagging for detection of endogenous protein levels in real time in a variety of cell lines, and easy implementation on the Agilent BioTek BioSpa 8 automated incubator and Synergy Neo2 hybrid multimode reader. Furthermore, we will demonstrate mechanistic characherization of ternary complex formation and ubiquitination using NanoBRET technology.
In this webinar, you will learn about:
• Understanding the importance of live-cell kinetic measurements of degradation and the ability to derive quantitative metrics for characterizing differences in efficacy among molecular glue analogs
• Using kinetic measurements to profile degradation selectivity of molecular glue across a panel of neo-substrates
• Combining kinetic measurements with NanoBRET technology for mechanistic validation and to relate ternary complex formation and /or ubiquination potency to the kinetic degradation profile
• How high throughput kinetic measurements of targeted protein degradation can be automated on the Agilent BioTek BioSpa 8 automated incubator and Synergy Neo2 hybrid multimode reader
