Wastewater is a challenging matrix for establishing consistent molecular biology methods. Since the COVID-19 pandemic, wastewater surveillance has been widely accepted as an economical and unbiased tool for surveillance of outbreaks and disease dynamics in a population. As a result, methods for wastewater sample preparation and PCR detection are constantly being improved and optimized. Here we will discuss the direct capture concentration and purification method in combination with digital PCR for quantifying SARS-CoV-2 levels in wastewater and tracking of variant circulation. We will also demonstrate how new data generated with improved methods can be matched to historical trending results.
